Perkin Elmer Operetta High Content Screening (HCS) system

The Operetta system is designed for high content screening (HCS) applications, combining the ability to capture images from large numbers of samples and the tools to generate unbiased, statistically valid data. Samples are loaded into the Operetta system using slides or glass-bottomed microplates including 96 and 386-well formats. Unsupervised automated image acquisition and autofocus aids reliable imaging with the minimum of hands-on time. Operetta is ideal for imaging cultured cells with incorporated CO2 and temperature environmental controls.

Spinning disc confocal technology

The Operetta system is fitted with a range of different objectives with a range of different magnifications allowing the combination of large fields of view followed by detailed analysis of specific areas of interest. Users also have the choice of high, numerical apertures or long working distance objectives. Fitted with a spinning disc confocal module, Operetta is capable of capturing high quality 2D and 3D images with higher contrast and more detail while at the same minimizing photobleaching and phototoxicity. Fast standard widefield fluorescence can also be used for routine applications.

Harmony: integrated acquisition and analysis software

Harmony is designed to make it easy to set up assays, automate experiments, analyze data and store, retrieve and present results. Its built in analysis tools makes it easy for biologists to turn their biology into unbiased quantitative data. It now incorporates ready-made solutions for a range of common applications to enable new users to get started with high content image analysis. STAR analysis provides a more detailed analysis of cellular morphology to accurately differentiate cellular phenotypes with parameters that describe cellular symmetry and compactness as well as the intracellular distribution of fluorescence. Texture analysis can be used where intensity and morphology are not suitable giving you a more complete picture of your biology. The PhenoLOGIC™ machine learning option enables you to teach the software to create optimized algorithms making segmentation and cell classification easier.

Columbus Image Data Storage and Analysis System

Handling the large amounts of data generated by high content screening and other large scale assays is tremendously challenging. Not only does the data need to be need to be stored safely and securely but also be easily accessible, analysable and available in a format that permits sharing with colleagues and comparison with other datasets. Columbus is a universal high-volume image data storage and analysis system that brings fast, secure access to images from a wide range of sources over a local network or the internet. The system is based on the OMERO, an open source development of the Open Microscopy Environment (OME) with the addition of an easy to use web-based interface. Columbus is compatible with data generated by virtually any imaging system thanks to the incorporation of the LOCI Bioformats plugin. It allows everyone in the lab to access, visualize and analyze image data from anywhere, regardless of the client operating system and without software installation. It includes powerful image analysis and secondary analysis tools, allows multiple samples, plates or batches to be compared for quality control. It is also includes the PhenoLOGIC™ machine-learning technology.
